Purpose

It is used for the selective isolation and identification of total coliform bacteria in drinking water and source water.

Inspection principle

Peptone, yeast powder and beef powder provide carbon and nitrogen sources, vitamins and minerals. Lactose is a fermentable sugar. AGAR is the coagulant of the culture medium. Dipotassium hydrogen phosphate is a buffer. Sodium sulfite can reduce alkaline fuchsin and cause it to fade. Bacteria that ferment lactose produce acetaldehyde. In the presence of alkaline fuchsia, acetaldehyde combines with sodium sulfite to form red colonies. Bacteria that rapidly ferment lactose produce a metallic luster, while those that do not ferment lactose form colorless and transparent colonies.

 

Usage

Weigh 52.5 grams of this product, add it to 1000mL of distilled water, heat to boiling, aliquot, and sterilize at 115℃ under high pressure for 20 minutes for later use.

 

Quality control

Prepare the culture medium according to the label usage, inoculate the following quality control strains, and place it in aerobic culture at 36±1℃ for 24±2 hours.
