Inspection principle
Pancreatic peptone provides carbon and nitrogen sources; Bile salt No. 3 inhibits Gram-positive bacteria, especially Gram-positive bacilli and Streptococcus faecalis. Lactose is a fermentable sugar. Potassium dihydrogen phosphate and potassium dihydrogen phosphate are buffers. Sodium chloride can maintain a balanced osmotic pressure. The glucuronidase contained in Escherichia coli acts on the β -glucuronidin bond of 4-Methylumbellifery-β -D-glucuronide (MUG for short), causing its hydrolysis. The released 4-methylumbellione produced blue-white fluorescence under a 366nm ultraviolet lamp. 97% of Escherichia coli, 10% of Salmonella and a small amount of Shigella have glucuronidase.
Usage
Weigh 37 grams of this product, add it to 1000mL of distilled water, heat and boil to dissolve, then aliquot into test tubes and sterilize at 115℃ under high pressure for 20 minutes for later use.
Quality control
Prepare the culture medium according to the label usage, inoculate the following quality control strains, and place it in an aerobic culture at 44.5±0.5℃ for 24-48 hours.
